Amezalpat builds momentum with both Fast Track and Orphan Drug Designations
BRISBANE, Calif., Feb. 10, 2025 (GLOBE NEWSWIRE) -- Tempest Therapeutics, Inc. (Nasdaq: TPST), a clinical-stage biotechnology company developing first-in-classi targeted and immune-mediated therapeutics to fight cancer, today announced that the U.S. Food and Drug Administration (FDA) has granted Fast Track Designation (FTD) to amezalpat (TPST-1120), an oral, small molecule, selective PPAR⍺ antagonist for the treatment of patients with hepatocellular carcinoma (HCC).

'This designation, following the Orphan Drug designation granted last month, reinforces the promise of amezalpat as a potential treatment option for patients affected by HCC. We look forward to working closely with the FDA and foreign regulatory agencies to develop amezalpat with the goal of bringing this promising therapy to patients.'
This is the second regulatory designation granted to amezalpat. The company announced in January that the U.S. FDA granted amezalpat with Orphan Drug Designation (ODD) following positive data across multiple key study efficacy and safety endpoints in a global randomized Phase 1b/2 clinical study evaluating amezalpat plus standard-of-care atezolizumab and bevacizumab versus atezolizumab and bevacizumab alone in the first-line treatment of patients with unresectable or metastatic HCC. Notable positive outcomes of the randomized comparison include a six-month improvement in median overall survival (OS) with a hazard ratio (HR) of 0.65 for patients receiving the amezalpat combination therapy. In addition, a survival benefit from the addition of amezalpat was preserved in key sub-populations including PD-L1 negative disease, which is consistent with amezalpat's proposed mechanism of action to target both the tumor cells directly and the patient's immune system.
About Hepatocellular Carcinoma
HCC is an aggressive cancer with rising mortality and is projected to become the third leading cause of cancer death by 2030.ii Every year, more than 900,000 people worldwide are diagnosed with HCC.iii Incidence and mortality are highest in East Asia and are increasing in parts of Europe and the US.iv In the US, HCC represents the fastest-rising cause of cancer-related death. iii

Amezalpat is an oral, small molecule, selective PPAR⍺ antagonist. Data suggests that amezalpat treats cancer by targeting tumor cells directly and by modulating immune suppressive cells and angiogenesis in the tumor microenvironment. In a global randomized phase 1b/2 study in first-line HCC patients, amezalpat in combination with atezolizumab and bevacizumab showed clinical superiority across multiple study endpoints, including overall survival in both the entire population and key subpopulations, when compared to patients receiving atezolizumab and bevacizumab alone, the standard of care. These randomized data were supported by additional positive results observed in the Phase 1 clinical trial in patients with heavily pretreated advanced solid tumors, including renal cell carcinoma and cholangiocarcinoma.

1 AI Super Stock Is Starting to Rebound, but Shares Still Look Cheap

Datadog stock remains more than 35% below the all-time high it reached four years ago. Many organizations are rapidly adopting AI-powered tools, which presents an opportunity for Datadog. The shares are trading near their cheapest price-to-sales ratio ever. 10 stocks we like better than Datadog › The rise of artificial intelligence (AI) is generating plenty of wealth on Wall Street -- and the winners won't be limited to just semiconductor stocks like Nvidia. Tech stocks across several subsectors will benefit, too. Let's take a look at one such stock, Datadog (NASDAQ: DDOG). Between 2019 and 2021, Datadog was one of the hottest names in the stock market. Shares advanced by more than 400% in only three years. However, as the stock market soured on tech stocks and speculative companies in 2022, Datadog shares plummeted. All told, the shares cratered by 68%, erasing the majority of their earlier gains. As of this writing, Datadog stock remains more than 35% off of the all-time high it touched in late 2021. Yet sentiment regarding the stock appears to have shifted. Datadog, which provides cloud monitoring services for enterprises, now boasts strong ratings from the analyst community. According to data compiled by Yahoo! Finance, there are 46 analysts covering Datadog. Of those, 10 rate it as a strong buy, 28 rate it a buy, and eight call it a hold. None of them rate it a sell or strong sell. Moreover, the average 12-month price target for Datadog shares is nearly $139. That's about 9% higher than the stock trades as of this writing. Datadog's business model is to sell monitoring services to organizations with significant cloud assets. This type of monitoring is critical to enterprises today, as operational downtime can result in serious consequences, including lost revenue, customer dissatisfaction, and even legal action. It already serves tens of thousands of clients across a range of industries, including e-commerce, gaming, and finance. While the type of monitoring that Datadog offers isn't new, what it is monitoring is changing. New large language models (LLMs) powered by AI algorithms have become much more important to organizations. Use of these models is rapidly spreading into the day-to-day operations of countless organizations. As this happens, their performance must be monitored, too. That has created a new source of revenue for Datadog, which is helping boost its growth. Consider the company's first-quarter results. Datadog noted that about 8.5% of its total revenue came from AI-native customers. That was up from 3.5% one year earlier -- showing meaningful growth for this new source of revenue. Management also raised its revenue guidance for the year by about $40 million, or 1%, on the back of this fast-growing new source of sales. Ultimately, these figures aren't game changers for Datadog, but they demonstrate that the AI revolution is benefiting the company. If it can continue to deliver on its higher guidance -- or even surpass it -- the stock should respond positively. In part, that's because Datadog's valuation remains near multiyear lows. Though Datadog's stock price has recovered significantly from its 2022 low point, its valuation -- as measured by its price-to-sales (P/S) ratio -- remains near the bottom of its range. As of this writing, Datadog shares trade at a P/S ratio of around 16. That's well below the peak levels above 60 that it reached in 2020 and 2021. It's also far below the stock's average of 28. Granted, a P/S ratio of 16 is still high compared to many stocks -- even within the tech sector. However, for long-term investors who want to establish a position in Datadog shares, it should be comforting that it doesn't appear to be overvalued. The stock appears to be rebounding after a steep decline. The AI revolution is playing a role in that comeback, and the analyst community is moderately bullish on the company's prospects. Finally, its current valuation is well below its long-term average. Overall, that suggests that this could be a good time for long-term investors to buy. Ovid's business model is centered on central nervous system (CNS) disorders, which are a challenging therapeutic area. Its pipeline includes novel compounds in early- to mid-stage development that target epilepsy and other genetically based neurological disorders. However, Wall Street analysts have called OVID a "Strong Buy," with all six analysts covering the stock rating it as such. Still, the company has also received a delisting notice from Nasdaq for failing to maintain the minimum bid price of $1.00 per share. Ovid has until Aug. 11, 2025, to regain compliance by maintaining a closing bid price of at least $1.00 for 10 consecutive trading days. Ovid's pipeline is small in size but very targeted. The most closely watched candidate is OV329, a highly selective small-molecule inhibitor of GABA aminotransferase (GABA-AT). OV329 is intended to treat epilepsy that has not responded to previous treatments. In preclinical models, the candidate has shown promise in controlling seizures while causing fewer side effects than existing treatments such as vigabatrin. Ovid Therapeutics anticipates Phase 1 trial results in the third quarter of 2025. The company has a good chance of its stock ticking upwards if the Phase 1 topline results for OV329 turn out positive. Ovid's other candidate is OV350, a next-generation KCC2 modulator that is also being tested in a Phase 1 trial. The company generated $130,000 in revenue from royalty agreements during Q1. At the end of the quarter, its cash, equivalents, and marketable securities totaled $43 million. This gives the company an estimated runway through the second half of 2026. Ovid has received a 'Strong Buy' rating on Wall Street because of its rich pipeline of rare disease treatments. The stock may appeal to long-term aggressive investors who are comfortable with volatility and have a strong interest in innovative science. However, I believe investors should wait until Ovid either clears the $1 delisting threshold or proves its clinical strategy with positive results. Analysts have an average target price of $2.88 for the stock, which implies upside potential of 860% from current levels. The Street-high price estimate stands at $4 for OVID stock. With a market cap of $609 million, Nuvation Bio is a clinical-stage biotech company that develops targeted oncology treatments for challenging cancers with limited treatment options. However, it may soon release a revenue-generating product to the market this year. NUVB stock is down 27.7% YTD, compared to the overall market gain. Nuvation Bio's pipeline includes programs addressing a variety of oncology targets, including breast, ovarian, and lung cancer. Its pipeline currently consists of four core programs: taletrectinib (ROS1 inhibitor), NUV-1511 (drug-drug conjugate), safusidenib (brain-penetrant IDH1 inhibitor), and NUV-868 (BET inhibitor). Taletrectinib, Nuvation's lead candidate for the treatment of ROS1+ and advanced non-small cell lung cancer, has already been approved in China and the United States. The company hopes to begin commercializing taletrectinib in mid-2025. In March, the firm secured $250 million in non-dilutive financing from Sagard Healthcare Partners, as well as $150 million in royalty financing and $50 million in debt, subject to approval. Nuvation also has the option to secure an additional $50 million in debt until June 30, 2026 if it completes its first commercial sale in the United States. Safusidenib is currently in Phase 2 trials, and NUV-1511 is in Phase ½ trials. The company intends to provide updates for both during the second half of 2025. Nuvation ended Q1 with $461.7 million in cash, cash equivalents, and marketable securities. A favorable FDA decision could open up significant market opportunities for the company. Nuvation Bio's late-stage pipeline, strong cash position, and clear commercialization path make it an appealing biotech growth stock to buy right now. However, the company's ability to generate profits after commercialization may take years, making it an appropriate choice for those with a high risk tolerance and a longer investment horizon. Overall, Wall Street rates NUVB stock a 'Strong Buy.' Of the seven analysts covering the stock, six rate Nuvation as a 'Strong Buy' while one recommends a 'Moderate Buy" rating. The average price target of $7.17 suggests that NUVB stock could rally around 258% over the next 12 months. The high price estimate of $10 suggests a gain of 400% over current levels.
