
Global NHS osteoarthritis study could lead to new treatments
More than 900 genetic links to osteoarthritis, including 500 which have never been reported, have been uncovered in a research study in Sheffield.Sheffield Teaching Hospitals NHS Foundation Trust said the "landmark" global study into osteoarthritis was the largest of its kind.The degenerative condition affects around 595 million people globally, and is the leading cause of disability and chronic pain worldwide, according to the hospital trust.The genome study used genetic information from nearly two million people, and was published in the journal Nature.
Led by Helmholtz Munich, the largest scientific research organisation in Germany, genetic codes and DNA of almost half a million osteoarthritis sufferers globally - including 1,000 volunteers from Sheffield - were compared with that of 1.5 million people without the disease, the trust said.The team identified 962 genetic variations more commonly found in those with osteoarthritis; more than 500 of which had not previously been identified, it said.The number osteoarthritis sufferers was predicted to rise to a billion people by 2050 and despite the impact "no disease-modifying treatments are currently available", researchers said.However the new work "paves the way for potential new drug treatments and personalised therapies," they continued.
'Repurpose existing treatments'
Prof Mark Wilkinson, Honorary Consultant Orthopaedic Surgeon at the trust and the University of Sheffield, said the findings were "hugely important" and would "open up exciting new avenues" for millions."As well as identifying potential drug targets and opportunities for us to repurpose existing treatments that already target these genes in other conditions, this research has also significantly advanced our understanding of the underlying biological mechanisms associated with the disease," Prof Wilkinson said.Eight biological processes which regulate the body's internal systems and cell function were found, "shedding light on the disease's biological mechanisms", the trust said.Some of these genes contain proteins already targeted by drugs approved for other conditions, which could accelerate treatment development, it added.

Country diary: What has the head of an ant, eyes of a housefly and legs of a spider?
I was standing on the verge beneath an oak tree when an insect dropped on to my bare arm. At first glance I thought it was a lacewing, but on closer inspection, something about it didn't look quite right. A quick Google search revealed it was an oak snakefly (Phaeostigma notata), a bizarre insect you've probably never heard of. They are rarely encountered and under-recorded, not because they're endangered, but because they're arboreal and tend to stay hidden high in the canopy, or tucked away in bark cracks and crevices. But wind or heavy rain, like the gusty downpour we'd had that morning, can send them to lower levels. It looked like a composite of insect body parts – a broad, flattened ant's head, a housefly's large compound eyes, spidery legs and damselfly-like wings with intricate venation. The presence of a formidable-looking long, needle‑like ovipositor indicated that this individual was female. Most unique was her elongated neck-like pronotum, which, when she reared up at the approach of my finger, gave her the appearance of a cobra poised to strike. It's this distinctive feature that gives these fantastical creatures their colloquial name. Snakeflies aren't flies (Diptera). They're related to the lacewings and antlions (Neuroptera), but are classified in a separate relictual order, Raphidioptera. They're considered living fossils, as they closely resemble species from the early Jurassic period, about 140m years ago. There are about 260 extant snakefly species worldwide, all found in temperate regions of the northern hemisphere, but only four have been recorded in the UK. Their strong mandibles seem at odds with a diet of soft-bodied prey, but they are voracious predators of aphids, mites and other small arthropods, their flexible pronotum enabling them to pursue prey into tight nooks and crannies. A recent stretch of warm weather, combined with high humidity, extended daylight hours and an abundance of succulent new growth, has created ideal conditions for an aphid population boom. Many trees in the locality have curled, distorted foliage – a telltale sign that sap-suckers have been piercing the leaf cuticle. But the oak was virtually unblemished, suggesting that the snakeflies are helping keep this damage in check. Australian moth uses starlight to travel thousands of miles

More info Three popular beaches in North Wales have become 'smoke-free zones' to safeguard nature and reduce the risk of wildfires. The initiative is only voluntary, with noticeboards erected at each site asking visitors not to light cigarettes. The beaches – Porth Dafarch and Cemlyn on Anglesey, and Porthor on the Llŷn Peninsula, Gwynedd – are all managed by National Trust Cymru. They were selected for their 'environmental significance' with support from RSPB Cymru. National Trust Cymru hopes smoke-free beaches will help protect wildlife and the coastlines by reducing environmental litter. According to Ash Wales Cymru, cigarette butts are one of the world's most littered items, posing a serious threat to marine environment and wildlife. Over time, said the charity, each butt will break up into microplastics, leaching toxic chemicals including lead, arsenic and formaldehyde into fragile marine ecosystems. It's hoped that by stopping people smoking on beaches, this will also protect young people from second-hand smoke and encourage them not to take up smoking. Backing the initiative - launched today on National Clean Air Day (June 19) - is Sarah Murphy, minister for mental health and wellbeing. She said: 'Our ambition is to create a smoke-free Wales by 2030 and creating more smoke-free spaces is one way to achieve this. "It is important we work together to limit the harmful effects of smoking as well as the associated health and environmental risks to our beautiful landscapes.' France has had similar bans since 2011, while stricter anti-smoking measures have been introduced in Italy and Greece. The latest North Wales additions are all renowned for the bird colonies and visitors. Cemlyn, on Anglesey's north coast, is home to an internationally important colony of Sandwich terns. Nesting colonies of Arctic and Common terns are also seen here, and, occasionally, the rarer Roseate tern. With around 1,500 pairs of terns counted in recent years, Cemlyn has the UK's third largest colony, with good water quality key to their prospects. Much of the surrounding coastline is a National Nature Reserve and a Site of Special Scientific Interest. Porth Dafarch, on Anglesey's Holy Island, has ancient rock formations that host a colony of choughs - the least common member of the crow family and recognised by its red bill and legs. Other visitors here include seals and dolphins. Dewi Davies, project manager for National Trust Cymru in North Wales, said: 'We work hard to look after these special places. "We are requesting that people who visit these beautiful locations help us to care for them by not smoking and disposing of harmful smoking materials. They could pose a risk to these special habitats and the wildlife that lives in them, or spark a wildfire.' The launch of the latest smoke-free spaces will be celebrated with a beach clean at Porth Dafarch on Thursday, June 19. Involving children from Ysgol Gynradd Kingsland, Holyhead, the event aims to highlight the dangers of cigarette waste on marine life. Smoke-free beaches are a partnership between the National Trust and ASH Wales. They believe there is strong public support for the idea in Wales. A national survey earlier this year showed 56% in favour of a smoking ban on beaches. A local survey in Anglesey showed even stronger support, with 87% in favour. Suzanne Cass, chief executive of ASH Wales, said: 'With 325,000 people in Wales still smoking, we must do everything we can to help them quit while creating a cleaner, healthier environment for all. "Expanding smoke-free outdoor spaces sends a clear message: smoking is not the norm - people, wildlife and the planet come first.'
