(NASDAQ:ENTA), a clinical-stage biotechnology company dedicated to creating small molecule drugs for viral infections and immunological diseases, today announced that the U.S. Food and Drug Administration (FDA) has approved a label expansion for MAVYRET® (glecaprevir/pibrentasvir), an oral pangenotypic direct acting antiviral (DAA) therapy. It is now approved as the only eight-week treatment for adults and pediatric patients three years and older with acute or chronic HCV infection without cirrhosis or with compensated cirrhosis.*

The label expansion was supported by data from the Phase 3, multicenter, single-arm prospective study evaluating the safety and efficacy of MAVYRET eight-week treatment in adults with acute HCV infection.1 The study results showed MAVYRET to be a highly efficacious treatment for people with acute HCV.1 The majority of the adverse events reported were mild or moderate in severity.1 The most common adverse events were fatigue, asthenia, headache, and diarrhea.1
The FDA granted Breakthrough Therapy Designation (BTD) for MAVYRET for the treatment of acute HCV. The BTD program is designed to expedite the development and review of medicines that are intended to treat a serious condition, and preliminary clinical evidence indicates that the drug may demonstrate substantial improvement over existing therapies on one or more clinically significant endpoints.3
HCV is a highly infectious blood-borne disease affecting the liver.2 If left untreated, HCV could lead to liver-related complications such as cirrhosis or liver cancer.2 Acute HCV refers to people recently infected with the virus and can be a short-term illness.4 However, for many people, acute infection leads to chronic infection.4 Current global clinical guidance call for the universal treatment of nearly all people with acute or chronic HCV infection.5 Widespread implementation of these guidelines has the potential to substantially reduce the global spread of the disease.5
Led by the World Health Organization, the public health community aims to eliminate HCV as a public health problem by 2030.6 This involves diagnosing and treating a large portion of those infected, as well as preventing new transmissions through measures like safe injection practices and harm reduction for people who inject drugs. However, approximately 80% of high-income countries, including the U.S., are not on track to achieve this goal until after 2050.7,8
About the Phase 3 M20-350 Study9The multicenter, single-arm prospective Phase 3 M20-350 clinical trial was designed to evaluate the safety and efficacy of MAVYRET® (glecaprevir/pibrentasvir) eight-week treatment in adults and pediatric patients with acute HCV infection. The study enrolled 286 treatment-naïve adult patients with acute HCV infection across 70 locations globally. Patients received oral tablets of MAVYRET® once daily for eight weeks and were followed for 12 weeks after the end of treatment. The primary endpoint was the percentage of patients with sustained virological response 12 weeks post-treatment (SVR12) in the Intention-to-Treat (ITT) population. Secondary endpoints included the percentage of patients achieving SVR12 in the Modified ITT-Virologic Failure (mITT-VF) population, and the percentage of patients with on-treatment virologic failure and post-treatment relapse in the ITT population. MAVYRET is a prescription medicine used to treat adults and children 3 years of age and older with:
Acute (recently infected) or chronic (lasting a long time) hepatitis C virus (hep C) genotypes 1, 2, 3, 4, 5 or 6 infection without cirrhosis or with compensated cirrhosis.
Hep C genotype 1 infection who have been previously treated with a regimen that contained a hep C NS5A inhibitor or an NS3/4A protease inhibitor, but not both.

What is the most important information I should know about MAVYRET?
Hepatitis B virus (hep B) reactivation: Before starting treatment with MAVYRET, your doctor will do blood tests to check for hep B infection. If you have ever had hep B infection, hep B could become active again during or after treatment for hep C with MAVYRET. Hep B that becomes active again (called reactivation) may cause serious liver problems, including liver failure and death. Your doctor will monitor you if you are at risk for hep B reactivation during treatment and after you stop taking MAVYRET.
Do not take MAVYRET if you:
Have moderate or severe liver impairment (Child-Pugh B or C) or any history of prior liver decompensation
Are taking the medicines atazanavir or rifampin
What should I tell my doctor before taking MAVYRET?
If you have had hep B infection, have liver problems other than hep C infection, have HIV-1 infection, have had a liver or a kidney transplant, and all other medical conditions.
If you are pregnant or plan to become pregnant, or if you are breastfeeding or plan to breastfeed. It is not known if MAVYRET will harm your unborn baby or pass into your breast milk. Talk to your doctor about the best way to feed your baby if you take MAVYRET.

On Wednesday, the FDA reported that five lots of Little Remedies Honey Cough Syrup, a 100% natural children's cough medicine for ages 1 and up, were voluntarily recalled by Medtech Products. The kid's cough syrup was found to be contaminated by Bacillus cereus, a bacterium that can cause two types of foodborne illnesses. Penumbra may gain market share in peripheral embolisation coils with Ruby XL
Penumbra has announced US Food and Drug Administration approval for its Ruby XL system, which includes three coils: the Ruby XL, the POD XL and the Packing Coil XL. The Ruby XL coil is a device used in vascular embolisation, a procedure to block or close off blood vessels, and can be used to frame large aneurysms, while the POD XL is designed for use in high-flow vessels and the Packing Coil XL is designed to adjust to the shape of any vessel up to 70cm in length. All Ruby XL system coils can be delivered through a 0.035-inch diagnostic catheter. The system is expected to enable Penumbra to increase its market share in peripheral embolisation coils, according to leading data and analytics company GlobalData. According to GlobalData's US healthcare facility invoicing database, Penumbra currently holds the majority market share for peripheral embolisation coils. Its new coil system allows for peripheral embolisation procedures to be done on a wider range of aneurysm sizes and in longer vessels. If the Ruby XL coil is widely adopted by physicians, Penumbra may be able to capture half the entire market. The approval of the new coil could put a bigger gap between Penumbra and its competitors in the market. Physicians may choose Ruby XL coils for their procedures in order to gain better control of large aneurysms. argenx Announces European Commission Approval of VYVGART Subcutaneous Injection for Chronic Inflammatory Demyelinating Polyneuropathy

VYVGART for SC injection is available as a vial or prefilled syringe and can be administered by a patient, caregiver, or healthcare professional. Treatment is initiated with a weekly dose regimen and may be adjusted to every other week based on clinical evaluation. "The EC's decision has been met with hope and enthusiasm by the European Patient Organisation for Dysimmune and Inflammatory Neuropathies (EPODIN). CIDP is a rare, debilitating, often progressive, immune-mediated neuromuscular disorder of the peripheral nervous system. The EC approval follows a positive recommendation from the Committee for Medicinal Products for Human Use (CHMP) and is based on positive results from the ADHERE clinical trial, the largest study of CIDP patients to date. In the study, 66.5% (214/322) of patients treated with VYVGART SC demonstrated evidence of clinical improvement, including in mobility, function and strength. Clinical benefit was seen across all patient subtypes, regardless of prior treatment. ADHERE met its primary endpoint (p<0.0001) demonstrating a 61% reduction (HR: 0.39 95% CI: 0.25; 0.61) in the risk of relapse versus placebo. 99% of trial participants elected to participate in the ADHERE open-label extension. The safety results were consistent with the known safety profile of VYVGART SC in previous clinical studies. About ADHERE The ADHERE trial was a multi-center, randomised, double-blind, placebo-controlled trial evaluating efgartigimod alfa SC for the treatment of CIDP. ADHERE enrolled 322 adult patients with CIDP, 130 of whom were based in Europe. The trial consisted of an open-label Stage A followed by a randomized, placebo-controlled Stage B. In order to be eligible for the trial, the diagnosis of CIDP was confirmed by an independent panel of experts. Patients entered a run-in stage, where any ongoing CIDP treatment was stopped and, in order to be eligible for Stage A, had to demonstrate active disease with clinically meaningful worsening on at least one CIDP clinical assessment tool, including INCAT, I-RODS, or mean grip strength. Treatment-naïve patients were able to skip the run-in period with proof of recent worsening. To advance to Stage B, patients needed to demonstrate evidence of clinical improvement (ECI) with efgartigimod alfa SC. ECI was achieved through improvement of the INCAT score, or improvement on I- RODS or mean grip strength if those scales had demonstrated worsening during the run-in period. In Stage B, patients were randomized to either efgartigimod alfa SC or placebo for up to 48 weeks. The primary endpoint was measured once 88 total relapses or events were achieved in Stage B and was based on the hazard ratio for the time to first adjusted INCAT deterioration (i.e. relapse). After Stage B, all patients had the option to roll-over to an open-label extension study to receive efgartigimod alfa SC.
