
Discovery of genes relevant to disease a ‘eureka moment'

Treatment for calcium pyrophosphate deposition (CPPD) disease — more commonly known as inflammatory arthritis — may soon take a major step forward following a major discovery by a University of Otago researcher.
There is an unmet need for treatment of CPPD disease, which focuses on the alleviation of inflammation with non-steroidal anti-inflammatory drugs.
But University of Otago microbiologist and immunologist Dr Tony Merriman has led an analysis of the genes of 550,000 Americans with the disease, while based at the University of Alabama, Birmingham.
It is a first-of-its-kind genome-wide association study into the disease and the research found two genes — RNF144B and ENPP1 — that cause the debilitating calcium pyrophosphate crystal deposits in joints.
He said the genes were detected both in people of European ancestry and African ancestry.
"The most significant result of our research was the discovery of one of the genes, ENPP1.
"The protein encoded by this gene controls the production of chemicals — adenosine monophosphate and inorganic pyrophosphate — that, together with calcium ions, lead to the formation of the CPP crystals," Dr Merriman said.
Little is known about the other gene — RNF144B — aside from the possibility it is involved in inflammation.
More was known about ENPP1 and of its potential importance to people with CPPD disease.
Drugs targeting the protein have been developed in the treatment of infectious disease and cancer and they could now be evaluated for the treatment of CPPD disease.
Dr Merriman said the discovery opened up promising new avenues for targeted prevention and treatment of CPPD disease, which were lacking at present.
Co-investigator and Brigham and Women's Hospital rheumatology, inflammation and immunity division researcher Dr Sara Tedeschi said the genome-wide association with ENPP1 was "particularly exciting" as a rheumatologist because "it makes sense".
"ENPP1 generates inorganic pyrophosphate, one of the components of CPP crystals.
"Patients with CPPD disease are desperate for an effective treatment and trials testing ENPP1 inhibitors in CPPD disease would be of great interest."
Dr Merriman said the researchers were "thrilled" about the potential impact of their discovery and the possibility of new drugs being developed for the treatment of CPPD disease.
"The findings of this study produced a 'eureka moment', which can be rare in a scientist's career."

The report said there was 'no radiation danger whatsoever'. An Iranian state television reporter, on live in the nearby town of Khondab, said the facility had been evacuated and there was no damage done to civilian areas around the reactor. Israel had warned earlier Thursday morning it would attack the facility and urged the public to flee the area. The Israeli military said Thursday's round of airstrikes targeted Tehran and other areas of Iran, without elaborating. It later said Iran fired a new salvo of missiles at Israel and told the public to take shelter. Israel's seventh day of airstrikes on Iran came a day after Iran's supreme leader rejected US calls for surrender and warned that any military involvement by the Americans would cause 'irreparable damage to them'. Israel also lifted some restrictions on daily life, suggesting the missile threat from Iran on its territory was easing. Already, Israel's campaign has targeted Iran's enrichment site at Natanz, centrifuge workshops around Tehran and a nuclear site in Isfahan. Its strikes have also killed top generals and nuclear scientists. A Washington-based Iranian human rights group said at least 639 people, including 263 civilians, have been killed in Iran and more than 1,300 wounded. In retaliation, Iran has fired some 400 missiles and hundreds of drones, killing at least 24 people in Israel and wounding hundreds. Heavy water helps cool nuclear reactors, but it produces plutonium as a byproduct that can potentially be used in nuclear weapons. That would provide Iran another path to the bomb beyond enriched uranium, should it choose to pursue the weapon. Iran had agreed under its 2015 nuclear deal with world powers to redesign the facility to relieve proliferation concerns. In 2019, Iran started up the heavy water reactor's secondary circuit, which at the time did not violate Tehran's 2015 nuclear deal with world powers. Britain at the time was helping Iran redesign the Arak reactor to limit the amount of plutonium it produces, stepping in for the US, which had withdrawn from the project after President Donald Trump's decision in 2018 to unilaterally withdraw America from the nuclear deal. The International Atomic Energy Agency, the United Nations' nuclear watchdog, has been urging Israel not to strike Iranian nuclear sites. IAEA inspectors reportedly last visited Arak on May 14. Due to restrictions Iran imposed on inspectors, the IAEA has said it lost 'continuity of knowledge' about Iran's heavy water production -- meaning it could not absolutely verify Tehran's production and stockpile. ADVERTISEMENT As part of negotiations around the 2015 deal, Iran agreed to sell off its heavy water to the West to remain in compliance with the accord's terms. Even the US purchased some 32 tons of heavy water for over $8 million (NZ$13.3 million) in one deal. That was one issue that drew criticism from opponents to the deal.

Otago rose to 197th in the rankings, following last year's placing of 214th, finishing with an overall score of 55.8. It was one of four New Zealand universities to improve its ranking. More than 1500 institutions in 106 countries are ranked, based on measures including a survey of academic reputation, staff to student ratios and citations of lecturers' research. The University of Auckland was New Zealand's highest-ranked institution in 65th place, with the other seven universities ranked between 197 and 410. Massey, Victoria and AUT also improved their rankings. Considered as a university system, New Zealand's universities performed well, the ranking said. "Among countries and territories with at least eight institutions featured in this year's rankings, New Zealand's overall average score of 51 puts the country fifth in the world for the overall quality of its higher education," it said. Only Hong Kong, the Netherlands, Sweden and Switzerland had better-performing university systems. The ranking placed Massachusetts Institute of Technology (MIT) first for the 14th consecutive year followed by Imperial College London and Stanford University. Other ranking systems last year placed New Zealand universities lower in their league tables.
