Intellia Therapeutics Announces Positive Three-Year Data from Phase 1 Trial of Lonvoguran Ziclumeran (lonvo-z) in Patients with Hereditary Angioedema (HAE) at the European Academy of Allergy and Clinical Immunology Congress
With up to three years of follow-up, a single dose of lonvo-z led to a 98% mean reduction in monthly HAE attack rate in all 10 patients
All 10 patients were attack-free and treatment-free for a median of 23 months through the latest follow-up, demonstrating the potential of lonvo-z to become the first one-time therapy for most HAE patients
Lonvo-z was well tolerated and continues to demonstrate a favorable safety profile
The global Phase 3 HAELO trial of lonvo-z has concluded screening ahead of schedule with more than half screened from U.S. sites; Intellia to provide an update on enrollment in the future
CAMBRIDGE, Mass., June 15, 2025 (GLOBE NEWSWIRE) — Intellia Therapeutics, Inc. (NASDAQ:NTLA), a leading clinical-stage gene editing company focused on revolutionizing medicine with CRISPR-based therapies, today announced three-year follow-up data from the Phase 1 portion of the ongoing Phase 1/2 study in patients with HAE after receiving a single dose of lonvoguran ziclumeran (lonvo-z, also known as NTLA-2002). Results were shared in an oral presentation at the European Academy of Allergy and Clinical Immunology (EAACI) Congress 2025, held June 13-16 in Glasgow, United Kingdom.
'Today's results underscore the promising potential of Intellia's approach to gene editing therapy – a one-time treatment that was well tolerated and offered a highly differentiated, durable effect for patients suffering from a serious disease,' said Intellia President and Chief Executive Officer John Leonard, M.D. 'Seeing all 10 patients in the Phase 1 portion of this study free from both HAE attacks and chronic therapy at nearly two years of median follow-up is incredibly encouraging. These data fuel our optimism for the outcomes of our ongoing Phase 3 HAELO study, which we expect to report in the first half of 2026, and highlight the strong value we believe it will offer patients, physicians and payers.'
'People living with HAE often report a reduced quality of life because they worry about the likelihood of their next attack, either because they still experience attacks or are reminded of it by their use of chronic therapy,' said Dr. Joshua Jacobs, Medical Director, Allergy and Asthma Clinical Research, Inc. 'Based on the data, it is reasonable to expect lonvo-z could offer patients the potential to be free from both physical HAE attacks and the burden of managing chronic HAE treatment.'
In the Phase 1 portion of the study, a one-time dose of 25 mg (N=3), 50 mg (N=4) or 75 mg (N=3) of lonvo-z was administered via intravenous infusion and plasma kallikrein protein levels were measured along with HAE attacks. At the time of the February 12 data cutoff, patients were attack-free and treatment-free for a median of nearly two years. With up to three years of follow-up, a single dose of lonvo-z led to a mean reduction in monthly HAE attack rate of 98% over the study period, compared to pre-treatment baseline. For all 10 patients, deep, dose-dependent and durable reductions in plasma kallikrein protein continued to be observed through the latest assessment.
Safety
Across all three dose levels, lonvo-z has been well tolerated and continues to demonstrate a favorable safety profile consistent with earlier data presented at EAACI in 2024. The most frequent adverse events during the study period were infusion-related reactions (IRRs). IRRs were mostly Grade 1 and resolved with all patients receiving the full dose. With up to 3 years of follow-up, no treatment-emergent serious adverse events were observed, and no treatment-related adverse events were observed during the period following 28 days after dosing.
Clinical Development Plans
Intellia's global Phase 3, randomized, double-blind, placebo-controlled HAELO trial is ongoing to assess the safety and efficacy of lonvo-z at the 50 mg dosage. The Company announced today the HAELO trial has successfully completed screening ahead of schedule, with over half of the patients being screened in the United States. The study is no longer recruiting and Intellia will provide an update on enrollment in the future. New and longer-term data from the Phase 2 portion of the ongoing Phase 1/2 study is planned to be presented in the second half of 2025. Intellia expects to submit a biologics license application (BLA) in 2026 to support the Company's plans for a U.S. launch in 2027. About the Lonvoguran Ziclumeran (lonvo-z, also known as NTLA-2002) Clinical Program
Intellia's ongoing Phase 1/2 study is evaluating the safety and efficacy of lonvo-z in adults with Type I or Type II hereditary angioedema (HAE). The Phase 1 portion of the study is an international, open-label study designed to identify the dose level of lonvo-z selected for further evaluation in the Phase 2 portion of the study. Enrollment in both portions of the Phase 1/2 study is complete. Intellia dosed the first patient in the global Phase 3, randomized, double-blind, placebo-controlled HAELO trial in January of 2025. Visit clinicaltrials.gov (NCT05120830) for more details.
About Lonvo-z
Based on Nobel Prize-winning CRISPR/Cas9 technology, lonvo-z has the potential to become the first one-time treatment for hereditary angioedema (HAE). Lonvo-z is an investigational in vivo CRISPR-based gene editing therapy designed to prevent HAE attacks by inactivating the kallikrein B1 ( KLKB1 ) gene, which encodes for prekallikrein, the kallikrein precursor protein. Interim Phase 1/2 clinical data showed dramatic reductions in attack rate, as well as consistent, deep and durable reductions in kallikrein levels. Lonvo-z has received five notable regulatory designations, including Orphan Drug and RMAT Designation by the U.S. Food and Drug Administration (FDA), the Innovation Passport by the U.K. Medicines and Healthcare products Regulatory Agency (MHRA), Priority Medicines (PRIME) Designation by the European Medicines Agency, as well as Orphan Drug Designation (ODD) by the European Commission.

National CineMedia (NASDAQ:NCMI) investors are sitting on a loss of 79% if they invested five years ago
Some stocks are best avoided. We really hate to see fellow investors lose their hard-earned money. Anyone who held National CineMedia, Inc. (NASDAQ:NCMI) for five years would be nursing their metaphorical wounds since the share price dropped 83% in that time. Furthermore, it's down 11% in about a quarter. That's not much fun for holders. While a drop like that is definitely a body blow, money isn't as important as health and happiness. Since shareholders are down over the longer term, lets look at the underlying fundamentals over the that time and see if they've been consistent with returns. This technology could replace computers: discover the 20 stocks are working to make quantum computing a reality. Because National CineMedia made a loss in the last twelve months, we think the market is probably more focussed on revenue and revenue growth, at least for now. Generally speaking, companies without profits are expected to grow revenue every year, and at a good clip. That's because fast revenue growth can be easily extrapolated to forecast profits, often of considerable size. Over five years, National CineMedia grew its revenue at 1.6% per year. That's not a very high growth rate considering it doesn't make profits. Nonetheless, it's fair to say the rapidly declining share price (down 13%, compound, over five years) suggests the market is very disappointed with this level of growth. We'd be pretty cautious about this one, although the sell-off may be too severe. We'd recommend focussing any further research on the likelihood of profitability in the foreseeable future, given the muted revenue growth. You can see below how earnings and revenue have changed over time (discover the exact values by clicking on the image). You can see how its balance sheet has strengthened (or weakened) over time in this free interactive graphic. It is important to consider the total shareholder return, as well as the share price return, for any given stock. Whereas the share price return only reflects the change in the share price, the TSR includes the value of dividends (assuming they were reinvested) and the benefit of any discounted capital raising or spin-off. It's fair to say that the TSR gives a more complete picture for stocks that pay a dividend. As it happens, National CineMedia's TSR for the last 5 years was -79%, which exceeds the share price return mentioned earlier. And there's no prize for guessing that the dividend payments largely explain the divergence! We're pleased to report that National CineMedia shareholders have received a total shareholder return of 19% over one year. That's including the dividend. There's no doubt those recent returns are much better than the TSR loss of 12% per year over five years. This makes us a little wary, but the business might have turned around its fortunes. While it is well worth considering the different impacts that market conditions can have on the share price, there are other factors that are even more important. Consider for instance, the ever-present spectre of investment risk. We've identified 1 warning sign with National CineMedia , and understanding them should be part of your investment process. York Water (NASDAQ:YORW) shareholders have endured a 26% loss from investing in the stock five years ago
The main aim of stock picking is to find the market-beating stocks. But every investor is virtually certain to have both over-performing and under-performing stocks. So we wouldn't blame long term The York Water Company (NASDAQ:YORW) shareholders for doubting their decision to hold, with the stock down 33% over a half decade. Now let's have a look at the company's fundamentals, and see if the long term shareholder return has matched the performance of the underlying business. AI is about to change healthcare. These 20 stocks are working on everything from early diagnostics to drug discovery. The best part - they are all under $10bn in marketcap - there is still time to get in early. In his essay The Superinvestors of Graham-and-Doddsville Warren Buffett described how share prices do not always rationally reflect the value of a business. One imperfect but simple way to consider how the market perception of a company has shifted is to compare the change in the earnings per share (EPS) with the share price movement. During the unfortunate half decade during which the share price slipped, York Water actually saw its earnings per share (EPS) improve by 2.6% per year. Given the share price reaction, one might suspect that EPS is not a good guide to the business performance during the period (perhaps due to a one-off loss or gain). Alternatively, growth expectations may have been unreasonable in the past. By glancing at these numbers, we'd posit that the the market had expectations of much higher growth, five years ago. Having said that, we might get a better idea of what's going on with the stock by looking at other metrics. Revenue is actually up 8.6% over the time period. A more detailed examination of the revenue and earnings may or may not explain why the share price languishes; there could be an opportunity. The graphic below depicts how earnings and revenue have changed over time (unveil the exact values by clicking on the image). It's probably worth noting that the CEO is paid less than the median at similar sized companies. It's always worth keeping an eye on CEO pay, but a more important question is whether the company will grow earnings throughout the years. So it makes a lot of sense to check out what analysts think York Water will earn in the future (free profit forecasts). When looking at investment returns, it is important to consider the difference between total shareholder return (TSR) and share price return. Whereas the share price return only reflects the change in the share price, the TSR includes the value of dividends (assuming they were reinvested) and the benefit of any discounted capital raising or spin-off. Arguably, the TSR gives a more comprehensive picture of the return generated by a stock. In the case of York Water, it has a TSR of -26% for the last 5 years. That exceeds its share price return that we previously mentioned. This is largely a result of its dividend payments! Investors in York Water had a tough year, with a total loss of 9.3% (including dividends), against a market gain of about 12%. However, keep in mind that even the best stocks will sometimes underperform the market over a twelve month period. Regrettably, last year's performance caps off a bad run, with the shareholders facing a total loss of 5% per year over five years. Generally speaking long term share price weakness can be a bad sign, though contrarian investors might want to research the stock in hope of a turnaround. While it is well worth considering the different impacts that market conditions can have on the share price, there are other factors that are even more important. Take risks, for example - York Water has 2 warning signs (and 1 which is significant) we think you should know about. If you are like me, then you will not want to miss this free list of undervalued small caps that insiders are buying. Weber Grill Restaurant to be first tenant in Orland Park downtown development
Weber Grill Restaurant will open a location in Orland Park next year, becoming the first tenant announced for the village's downtown development project northwest of La Grange Road and 143rd Street. The company is owned by the same family that makes the grills, which are used to prepare meals at the restaurants. Weber Grill will build an 8,500-square-foot space at the southwest corner of La Grange and 142nd Street, said Ramzi Hassan, president of Edwards Realty Co. Orland Park-based Edwards was selected by the village to develop that has been called the Main Street Triangle project. 'This is exactly the type of tenant we envisioned when we first imagined downtown Orland Park,' Hassan said in a news release announcing the restaurant's plans. 'Weber Grill represents a high-quality, experience-driven restaurant that aligns perfectly with our vision to create a dynamic, walkable destination for the community.' Planned to open next spring, the restaurant will feature indoor and outdoor seating, as well as a hands-on cooking school offering both public and private cooking classes, according to Weber Grill Restaurants. The company also has restaurants in downtown Chicago and suburban locations in Lombard and Schaumburg. 'We're thrilled to bring the unique Weber Grill experience to the new downtown Orland Park,' Jon Crost, chief operating officer of Weber Grill Restaurants, said in a news release. 'This location allows us to connect with a vibrant and growing community while being part of a visionary redevelopment that emphasizes both quality and community.' Hassan said Friday that Weber 'is a really good brand that's a household name,' and its decision to open in Orland Park could attract other tenants. He said that Weber was looking to the Orland Park market for a restaurant, but 'they were more interested in the type of development they are joining.' 'They had to find the right fit,' Hassan said. He said Edwards has been very selective about the users and businesses coming in to the development, and said the Weber Grill Restaurant could be destination drawing customers from beyond the Orland Park area. 'It's not like they have 300 restaurants all over the country,' Hassan said. Edwards plans to develop more than 140,000 square feet of entertainment, retail and office space. No residential space is planned. An expansion of Crescent Park, to be renamed Heroes Park, is included, and what is now a stormwater detention pond in the northeast corner of the triangle would be enhanced with fountains. The company and village held a ceremonial groundbreaking for the downtown project in March. Edwards owns and operates Orland Park Crossing, a retail center northeast of La Grange and 143rd, directly to the east of the downtown development. The downtown development would be situated around the Ninety7Fifty on the Park apartments, University of Chicago Medicine Center for Advanced Care, a parking garage and the 143rd Street Metra station. Hassan said he expects Edwards to break ground in spring on space for a second tenant, which would be on a site west of La Grange on the south side of 142nd.
